| Function | Human interleukin-18 (IL-18) is a pro-inflammatory cytokine with roles in both innate and adaptive immunity. IL-18 is primarily expressed by macrophages and monocytes as the inactive precursor (pro-IL-18), whereby the biologically active IL-18 results from Caspase-1 cleavage of the pro-domain. IL-18 stimulates natural killer (NK) cells and T-helper 1 (Th1) cells. IL-18 signals through a receptor complex made up of IL-18R alpha, the primary binding subunit which recruits IL-18R beta to form a high-affinity signaling complex which leads to the recruitment of the adaptor protein, MyD88. MyD88 then facilitates the activation of interleukin-1 receptor-associated kinases (IRAKs) and TNF receptor-associated factor 6 (TRAF6). IL-18 signaling is often potentiated by other cytokines, particularly IL-12, which together amplify the production of IFN-gamma. |
