Function | Human interleukin-18 (IL-18) is a pro-inflammatory cytokine with roles in both innate and adaptive immunity. IL-18 is primarily expressed by macrophages and monocytes as the inactive precursor (pro-IL-18), whereby the biologically active IL-18 results from Caspase-1 cleavage of the pro-domain. IL-18 stimulates natural killer (NK) cells and T-helper 1 (Th1) cells. IL-18 signals through a receptor complex made up of IL-18R alpha, the primary binding subunit which recruits IL-18R beta to form a high-affinity signaling complex which leads to the recruitment of the adaptor protein, MyD88. MyD88 then facilitates the activation of interleukin-1 receptor-associated kinases (IRAKs) and TNF receptor-associated factor 6 (TRAF6). IL-18 signaling is often potentiated by other cytokines, particularly IL-12, which together amplify the production of IFN-gamma. |
Tissue Specificity | IL-18 expression has been reported in various tissues and cell types, particularly monocytes and macrophages, dendritic cells (DCs), liver (Kupffer cells), spleen, brain (microglia), placenta, epithelial cells (gastrointestinal tract, lung), skin (keratinocytes), endothelia cells (vascular tissue), and adipocytes. |
