Background
Alternative Names | Adiponectin, Acrp30, Adipocyte complement-related 30 kDa protein, Gelatin-binding protein, GBP28, Adipose most abundant gene transcript 1 protein, APM1, C1Q and collagen domain containing |
Function | Adiponectin (Acrp30) is an adipokine secreted by adipocytes enhancing insulin sensitivity in peripheral tissues by promoting glucose uptake in skeletal muscle while suppressing gluconeogenesis in the liver in its role regulating metabolic processes and maintaining energy homeostasis. Acrp30 facilitates fatty acid oxidation in muscles and reduces lipid accumulation in the liver. This action helps lower circulating levels of free fatty acids, contributing to overall lipid balance. Adiponectin exerts anti-inflammatory effects by inhibiting pro-inflammatory cytokines. Adiponectin forms multimeric complexes. These include low molecular weight trimers, medium molecular weight hexamers, and high molecular weight (HMW) oligomers. The HMW is considered the most biologically active in improving insulin sensitivity. A truncated form of Adiponectin composed of the C-terminal globular domain has been termed gAdiponectin. Adiponectin exerts its effects mainly through two receptors. AdipoR1 is predominantly expressed in skeletal muscle, it has a high affinity for the gAdiponectin. AdipoR2 is mainly expressed in the liver, it binds both full-length and gAdiponectin. |
Tissue Specificity | Adiponectin is reported primarily in the mature adipocytes in white adipose tissue, with lower expression reported in brown adipose tissue and the placenta. Preadipocytes express low to negligible levels of Adiponectin, with significant upregulation reported following differentiation into mature adipocytes. |
Cellular Localization | Secreted |
Involvement in Disease | Dysregulation of Adiponectin has been reported in metabolic disorders (obesity, type 2 diabetes, nonalcoholic fatty liver disease [NAFLD]), cardiovascular diseases (atherosclerosis, coronary artery disease, hypertension), polycystic ovary syndrome (PCOS), and cancer (breast, endometrial, colon). |
