Background
Alternative Names | CCL17, TARC, ABCD-2, SCYA17, C-C Motif Chemokine 17, Small-inducible Cytokine A17, T cell-directed CC chemokine, Thymus and Activation-regulated Chemokine |
Function | TARC, also known as, CCL17, is a member of the CC chemokine family primarily functioning in a chemotactic role with T lymphocytes (especially Th2 cells), dendritic cells, and regulatory T cells, toward sites of inflammation or lymphoid tissues. TARC binds to chemokine receptor CCR4, a G protein-coupled receptor (GPCR), which is expressed on immune cells including Th2 cells, regulatory T cells, and some dendritic cell subsets. Upon binding TARC, CCR4 undergoes a conformational change that activates intracellular heterotrimeric G proteins leading to inhibition of Adenylyl Cyclase and activation of MAP kinases (ERK1/2) to drive chemotaxis. |
Tissue Specificity | TARC is expressed in various tissues related to its role in immune responses, such as thymus tissue, lymph nodes, spleen, skin (Langerhans cells), lung tissue (bronchial epithelial cells), liver tissue, kidney tissue and the gastrointestinal tract. |
Cellular Localization | Secreted protein |
Involvement in Disease | Dysregulation of TARC is implicated in allergy (atopic dermatitis and asthma), autoimmune disorders (rheumatoid arthritis and inflammatory bowel disease), and Hodgkin lymphoma. |
