Function | Human Interleukin-10 (IL-10) is a multifunctional cytokine with potent anti-inflammatory and immunoregulatory roles, as well as stem cell biology and tissue homeostasis. IL-10 suppresses inflammatory responses by downregulating the expression of pro-inflammatory cytokines (like TNF-alpha, IL-6, and IL-1 beta), chemokines, and co-stimulatory molecules produced by antigen-presenting cells (APCs) such as macrophages and dendritic cells. IL-10 influences T cell responses by promoting the development and function of regulatory T cells (Tregs), which are crucial for maintaining immune tolerance. Additionally, it can inhibit the antigen-presenting function of dendritic cells, thereby dampening the activation of effector T cells and curtailing prolonged immune reactions or autoimmune disorders. IL-10 exerts its effects by binding to a heterodimeric receptor composed of IL-10 receptor 1 (IL-10R1) and IL-10 receptor 2 (IL-10R2). IL-10R1 is primarily responsible for high-affinity binding to IL-10, while IL-10R2 is essential for signal transduction via the JAK1/Tyk2-STAT3 signaling cascade and is shared with other cytokine receptors. |
Tissue Specificity | IL-10 expression is reported in Macrophages, Monocytes, Dendritic Cells (DCs), Regulatory T Cells (Tregs), some populations of Th1 and Th2 cells, as well as Tr1 cells. Expression has also been reported in epithelial tissue (gut, respiratory tract, and skin), the placenta and trophoblasts, astrocytes and microglia, lymphoid tissue (spleen, thymus, and lymph nodes) and liver tissue. |
