Function | Interleukin-12 (IL-12) is a cytokine bridging innate and adaptive immunity, exerting effects on immune responses, particularly in the context of intracellular pathogens and tumor surveillance. IL-12 is instrumental in driving the differentiation of naïve CD4⁺ T cells into T helper type 1 (Th1) cells. IL-12 further stimulates these cells, as well as natural killer (NK) cells, to produce interferon-gamma (IFN-gamma). Additionally, IL-12 supports the activation and proliferation of cytotoxic T lymphocytes (CTLs). IL-12 is a disulfide-linked heterodimer composed of a 40 kD cytokine subunit encoded by the IL12B gene, and a 35 kD subunit encoded by the IL12A gene. The IL-12 receptor is a heterodimer composed of two subunits: IL-12R beta1 and IL-12R beta2. These subunits are expressed on the surface of T cells and NK cells, where the JAK/STAT signaling pathway—especially STAT4 activation—translates extracellular signals into effective immune responses. |
Tissue Specificity | Human IL-12 is chiefly expressed by antigen-presenting cells (APCs)—especially dendritic cells and macrophages—with B cells contributing under specific conditions. These cells are primarily located in secondary lymphoid organs (lymph nodes and spleen) contributing to activities to prime adaptive immune responses but may be found in peripheral tissues where local immune reactions occur (skin, lungs, gastrointestinal tract). |
