Function | IL-17A is a pro-inflammatory cytokine primarily produced by T helper 17 (Th17) cells, as well as by other immune cells such as gamma-delta T cells and group 3 innate lymphoid cells (ILC3s). The functional homodimer exerts its effects by binding to a receptor complex composed of the IL-17RA and IL-17RC subunits. Upon ligand binding, the receptor recruits the adaptor protein ACT1 (also known as CIKS). The ACT1 adaptor facilitates the activation of the NF-kappa B pathway, leading to the transcription of numerous pro-inflammatory genes. In addition to NF-kappa B, IL-17A signaling triggers MAPK cascades (such as ERK, JNK, and p38 pathways), which further amplify the inflammatory response via inflammatory cytokines and chemokines, such as IL-6 and IL-8. |
Tissue Specificity | IL-17A is expressed by several immune cell types—most notably Th17 cells, gamma-delta T cells, and ILC3s—and is predominantly found in secondary lymphoid organs, mucosal surfaces (such as the gut, lung, and skin), and sites of inflammation. |
