Background
Alternative Names | Interleukin-8, IL8, Granulocyte chemotatic protein 1, GCP1, C-X-C motif chemokin 8, LECT, LUCT, LYNAP, T cell chemotactic factor, TCF, TSG1, Neutrophil-activating protein 1,MDNCF, MONAP, NAF, NAP1, NCF |
Function | IL-8 (CXCL8) is a chemokine that plays a pivotal role in orchestrating the innate immune response and inflammation. It is primarily involved in recruiting and activating neutrophils to sites of infection or tissue damage. IL-8 is secreted by a range of cells—including macrophages, endothelial cells, and epithelial cells—in response to inflammatory stimuli. Upon release, IL-8 binds to its G protein-coupled receptors, CXCR1 and CXCR2, on target cells, triggering signaling pathways that promote chemotaxis, degranulation, and the respiratory burst in neutrophils. Beyond its role in immune cell recruitment, IL-8 also contributes to angiogenesis and tissue remodeling, processes that are important in wound healing and may influence tumor progression. |
Tissue Specificity | IL-8 expression has been reported in macrophages, epithelial, endothelial cells, and fibroblasts. |
Cellular Localization | Secreted |
Involvement in Disease | Dysregulation of IL-8 is implicated in inflammatory disorders such as rheumatoid arthritis, psoriasis, and inflammatory bowel disease, in addition to acute inflammatory states such as sepsis and acute respiratory distress syndrome (ARDS), where increased IL-8 drives excessive recruitment and activation of neutrophils, contributing to tissue damage. IL-8 is also implicated in Chronic lung conditions, including cystic fibrosis and chronic obstructive pulmonary disease (COPD); and atherosclerosis and other vascular inflammatory disorders. IL-8 promotes tumor growth, angiogenesis, and metastasis by shaping the tumor microenvironment and has been reported in various malignancies—including lung, colon, prostate, and breast cancers. |
