Function | Matrix Metalloproteinase-2 (MMP-2) is a zinc protease involved inextracellular matrix (ECM) remodeling. Structurally, MMP-2 consists of a signal peptide, inhibitory pro-domain, catalytic domain (with the zinc-binding motif and fibronectin repeats), hinge region, and a regulatory hemopexin-like C-terminal domain. MMP-2 is secreted as an inactive proenzyme (proMMP-2). Activation of MMP-2 involves cleavage of the pro-domain mediated at the cell surface by membrane-type metalloproteinases (MT-MMPs), particularly MT1-MMP (MMP-14). Once activated, MMP-2’s activity is tightly regulated by TIMPs (especially TIMP-2) to prevent excessive ECM degradation. |
Tissue Specificity | Matrix metalloproteinase‐2 (MMP‐2) is widely expressed in various tissues and cell types. MMP-2 expression is reported in macrophages, connective tissue (fibroblasts, myofibroblasts), vascular tissue (endothelial cells, smooth muscle cells), epithelial tissue, cardiac tissue (cardiac myofibroblasts, myocytes), central nervous system (CNS) tissue (neurons, astrocytes), placenta tissue, bone and cartilage (osteoblasts, chondrocytes), and cancer tissue (tumor and stromal cells). |
