| Function | Matrix Metalloproteinase-2 (MMP-2) is a zinc protease involved inextracellular matrix (ECM) remodeling. Structurally, MMP-2 consists of a signal peptide, inhibitory pro-domain, catalytic domain (with the zinc-binding motif and fibronectin repeats), hinge region, and a regulatory hemopexin-like C-terminal domain. MMP-2 is secreted as an inactive proenzyme (proMMP-2). Activation of MMP-2 involves cleavage of the pro-domain mediated at the cell surface by membrane-type metalloproteinases (MT-MMPs), particularly MT1-MMP (MMP-14). Once activated, MMP-2’s activity is tightly regulated by TIMPs (especially TIMP-2) to prevent excessive ECM degradation. |
