Background
Alternative Names | Neurotrophin 3, NT3, NTF3, Nerve Growth Factor 2, NGF2, HDNF |
Function | Neurotrophin 3 (NT-3) belongs to the neurotrophin family and primarily functions as a growth factor that promotes the survival, growth, and differentiation of neurons and oligodendrocytes. NT-3 stimulates the growth and branching of axons and dendrites. This process is crucial for the establishment of neuronal circuits during development and for the regeneration of neural connections after injury. NT-3 enhances neuroprotection and regulates synaptic plasticity in mature neurons. Outside of the CNS, NT-3 promotes differentiation and survival of peripheral, motor, and sensory neurons. NT-3 primarily binds to TrkC as the high affinity signaling receptor, but association with TrkB and the low-affinity p75-NTR has been reported.
In cell culture, NT-3 has been used to maintain neuronal cells in culture, the generation of peripheral neurons and oligodendrocyte precursors. It has also been used to differentiate human pluripotent stem cells (hPSCs) to glutamatergic and GABAergic neurons and the culture of 3-D organoids. |
Tissue Specificity | NT-3 expression has been identified in brain (cerebral cortex, hippocampus, cerebellum), spinal cord, peripheral nerves, ganglia, and to a lesser extent in skin, skeletal muscles, heart, kidney, and liver. |
Cellular Localization | Secreted protein |
Involvement in Disease | Dysregulation of NT-3 is associated with Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, diabetic neuropathy, Charcot-Marie-Tooth disease, depression, anxiety, amyotrophic lateral sclerosis (ALS), and muscular dystrophies. |
