Determined by a cell proliferation assay using mouse Balb/c 3T3 cells. The ED50 for this effect is typically 1-3 ng/mL | Recombinant Human PDGF-BB (2 ug/lane) on SDS-AGE under reducing (R) and non-reducing (NR) conditions. The gel was stained using Coomassie® Blue showing single bands at 13 kDa and 28 kDa, respectively, and purity greater than 96%. |
Background
Alternative Names | Platelet Derived Growth Factor BB, PDGFBB, Platelet-Derived Growth Factor B, PDGFB, PDGF Subunit B, Epididymis Secretory Sperm Binding Protein, Oncogene SIS, SIS, Proto-Oncogene C-SIS, C-SIS, Becaplermin |
Function | Platelet-Derived Growth Factor BB (PDGF-BB) is a protein with roles in regulating of many processes including embryonic development, angiogenesis, cell proliferation and differentiation. PDGF-BB acts on mesenchymal cells as a mitogen for connective tissue formation, particularly fibroblasts, vascular smooth muscle cells and glial cells. PDGF-BB is present in the alpha granules of platelets. As such, PDGF-BB promotes angiogenesis, chemotaxis of fibroblasts and collagen synthesis in tissue repair. Additionally, PDGF-BB supports development of kidney glomeruli, the differentiation of neural stem cells and has neuroprotective functions on mature neurons during injury. PDGF-BB primarily binds to the PDGFR beta (CD140B) homodimer, and with lower affinity to the PDGFR alpha (CD140a) homodimer or PDGFR alpha/PDGFR beta heterodimer leading to activation of the MAPK, PI3K, and STAT3 pathways. |
Tissue Specificity | PDGF-BB expression has been identified in endothelial tissue, smooth muscle tissue, heart, lung, brain, and neurons. |
Cellular Localization | Secreted protein |
Involvement in Disease | Dysregulation of PDGF-BB is associated with atherosclerosis, pulmonary hypertension, glomerulonephritis, diabetic retinopathy, keloids, hypertrophic scars, cancer (glioma, sarcoma, and tumor angiogenesis). |
