Function | Roof Plate-specific Spondin 1 (R-Spondin 1 or RSPO1) is a key regulator of Wnt signaling with diverse functions in during embryonic development, tissue homeostasis, and stem cell maintenance. Its precise role depends on the cellular context and interactions with other signaling pathways. RSPO1 enhances Wnt signaling by competing with Wnt antagonist DKK-1 for binding to LRP6 (Low-Density Lipoprotein Receptor-related Protein 6) and inhibiting its interaction with the co-receptor Kremen. This inhibition leads to the stabilization of the Frizzled/LRP6 receptor complex, which initiates downstream signaling events mediated by Wnt proteins. The role in tissue patterning is observed in expression of RSPO1 in the roof plate boundary during early development leading to involvement in organogenesis and gonadal development of both ovaries and testes. As the in vivo roles of RSPO1 have been elucidated in tissue patterning and stem cell maintenance, RSPO1 plays a more common role for in vitro cell culture and the growth and maintenance of 3-D organoid cultures. |
