Function | Roof plate-specific Spondin 3 (R-Spondin 3 or RSPO3) is a key regulator of Wnt signaling with diverse functions in during embryonic development, tissue homeostasis, and stem cell maintenance. In embryonic development, RSPO3 is involved in various processes, including limb bud development, skeletal patterning, and organogenesis. In adults, it plays a role in maintaining tissue homeostasis, particularly in stem cell niches where Wnt signaling is critical for the regulation of stem cell self-renewal and differentiation. RSPO3 enhances Wnt signaling by competing with Wnt antagonist DKK-1 for binding to Low-Density Lipoprotein Receptor-related Proteins LRP4, LRP5 or LRP6 and inhibiting its interaction with the co-receptor Kremen. This inhibition leads to the stabilization of the Frizzled/LRP receptor complex, which initiates downstream signaling events mediated by Wnt proteins. As the in vivo roles of RSPO3 have been elucidated in tissue patterning and stem cell maintenance, RSPO3 plays a more common role for in vitro cell culture and the growth and maintenance of 3-D organoid cultures. |
