Background
Alternative Names | Tumor Necrosis Factor alpha, TNF, TNFA, APC1, TNFSF2, Tumor Necrosis Factor Ligand Superfamily Member 2, Differentiation Inducing Factor, DIF, Cachectin |
Function | Tumor Necrosis Factor-alpha (TNF-alpha) was initially associated as a proinflammatory cytokine expressed by macrophages. However, TNF-alpha also regulates apoptosis, tumor cell death, lipid metabolism and cell differentiation of T cells, B cells and NK cells. There are 2 receptors for TNF-alpha, a ubiquitously expressed TNFR1 (CD120a) homotrimer containing an apoptotic pathway trigger and the TNFR2 (CD120b) homotrimer that is commonly restricted to epithelial, endothelial and specific immune cells. |
Tissue Specificity | TNF-alpha expression has been identified in adipose tissue, microglia, fibroblasts, endothelia tissue, and epithelial tissue. |
Cellular Localization | Membrane-bound and cleaved soluble protein forms |
Involvement in Disease | Dysregulation of TNF-alpha is implicated in rheumatoid arthritis, inflammatory bowel disease (IBD), psoriasis, ankylosing spondylitis, systemic lupus erythematosus (SLE), systemic sclerosis (SSc), multiple sclerosis (MS) and insulin resistance in type 2 diabetes. |
